Cytosolic Distribution of Cd2+, Cu2+, and Zn2+ Among Metallothionein-Like Proteins in Lobster Hepatopancreas

چکیده
Zinc and Cu metabolism in crustacea changes dramatically during the molt cycle. Metallothionein (MT) and MT-like proteins are implicated in the handling of both metal ions as well as in the detoxification of Cd. Distribution of Cd2 / Cu / and Zn2 / in cytosol and among MT-like species has been determined. Lobsters were treated to induce MT as in mammals with dexamethasone (dex) or stress from the surgical removal of a small walking toe both with and without a subsequent injection of CdCI2. Untreated tissue contains only Cu-MTII and no other significant amounts of metal-bound MTs. Exposure to dex or surgery alone causes no change in the MTs. Exposure to Cd retains the Cu-MTII and produces little Cd-MTI,II. When dex or surgery is combined with Cd2 / injection, Cu-MTII is unaffected and large quantities of Cd-MTI,II are made. Preliminary results indicate that dex or surgery induces the synthesis of apo-MT in the hepato-pancreas which serves as a site for binding cadmium.

Exposure of human proximal tubule cells to cd2+, zn2+, and Cu2+ induces metallothionein protein accumulation but not metallothionein isoform 2 mRNA.
The organization of the human metallothionein (MT) gene family is more complex than the commonly used mouse and rat models. The human MTs are encoded by a family of genes consisting of 10 functional and 7 nonfunctional MT isoforms. One objective of this study was to determine if the accumulation of MT protein in cultures of human proximal tubule (HPT) cells exposed to metals is similar to that ...
متن کاملDifferential Responses of Groel and Metallothionein Genes to Divalent Metal Cations and the Oxyanions of Arsenic in the Cyanobacterium Synechococcus Sp. Strain Pcc7942
A better understanding of the resistance mechanisms used by the cyanobacterium Synechococcus sp. PCC 7942 could guide attempts to develop this organism for use in bioremediation. This bacterium exhibits extraordinary resistance to arsenate. Possible mechanisms include exclusion from the cell, intraand extra-cellular binding, and potential interactions with proteins such as metallothioneins, whi...
متن کاملZinc Supplementation Protects against Cadmium Accumulation and Cytotoxicity in Madin-Darby Bovine Kidney Cells
Cadmium ions (Cd2+) have been reported to accumulate in bovine tissues, although Cd2+ cytotoxicity has not been investigated thoroughly in this species. Zinc ions (Zn2+) have been shown to antagonize the toxic effects of heavy metals such as Cd2+ in some systems. The present study investigated Cd2+ cytotoxicity in Madin-Darby bovine kidney (MDBK) epithelial cells, and explored whether this was ...
متن کاملDifferential expression of human metallothionein isoform I mRNA in human proximal tubule cells exposed to metals.
In contrast to the single metallothionein (MT)-1 gene of the mouse, the human MT-1 gene family is composed of seven active genes and six pseudogenes. In this study, the expression of mRNA representing the seven active human MT-1 genes was determined in cultured human proximal tubule (HPT) cells under basal conditions and after exposure to the metals Cd2+, Zn2+, Cu2+, Hg2+, Ag2+, and Pb2+. Basal...
متن کاملExpression of BjMT2, a metallothionein 2 from Brassica juncea, increases copper and cadmium tolerance in Escherichia coli and Arabidopsis thaliana, but inhibits root elongation in Arabidopsis thaliana seedlings.
The protective function of a plant type-2 metallothionein was analysed after expression in Escherichia coli and in Arabidopsis thaliana seedlings. BjMT2 from Brassica juncea was expressed in E. coli as a TrxABjMT2 fusion protein. After affinity chromatography and cleavage from the TrxA domain, pure BjMT2 protein was obtained which strongly reacted with the thiol reagent monobromobimane. Escheri...
